There is a growing interest in addressing erectile dysfunction (ED) with tadalafil (Cialis®). Sildenafil (100 mg) and tadalafil (60 mg) were administered to 30 male Wistar rats (12 weeks old) randomly divided into three groups: the standard sildenafil group (n=10), the tadalafil+tadalafil-PDE5i (20 mg/kg/day) group (n=10), and the tadalafil+Cialis® (60 mg/kg/day) group (n=10). The results demonstrated that sildenafil significantly enhanced arterial blood flow, whereas tadalafil significantly reduced blood flow in the tadalafil+Cialis® group. Furthermore, tadalafil-induced vasoconstriction was attenuated by the PDE5i, suggesting a role for PDE5i in the vasoconstriction response to tadalafil in ED patients.

Materials and Methods:

A total of 20 male Wistar rats (18 weeks old) were divided into three groups. The standard sildenafil group (n=10) and the tadalafil+tadalafil-PDE5i (20 mg/kg/day) group (n=10) were administered daily for 36 weeks. Blood samples were taken from each animal at baseline and after 12 weeks of treatment with sildenafil, tadalafil, or Cialis®. The animals were euthanized at the end of the experimental period, and aortic rings were removed and washed in tap water. Sildenafil and tadalafil-PDE5i were dissolved in saline, and blood samples were drawn at baseline and after 12 weeks of treatment. Vascular reactivity was determined by hematoxylin and eosin staining. Vascular and vascular endothelial function was assessed by a microangiography (MEG) using a rat cardiac catheterization system (Model B100, model 910, Wetzlar, Germany). The results showed that sildenafil significantly increased the mean arterial pressure (MAP) in the arterial wall of the tadalafil+tadalafil-PDE5i group compared to the control group (3.4 mmHg and 3.5 mmHg, respectively). There was a significant difference between the groups (P<0.05). The results demonstrated that tadalafil-induced vasoconstriction was attenuated by PDE5i, suggesting a role for PDE5i in the vasoconstriction response to tadalafil in ED patients.

The right arterial blood pressure was measured by an intra-arterial pressure monitoring syringe (Model 910, model, Wetzlar, Germany). A total of 20 rats in each group were used in this study. Results:

No significant differences in the values of the variables, such as systolic arterial pressure, diastolic arterial pressure, and mean arterial pressure at 12 weeks of treatment, were observed between the tadalafil+Cialis® and the standard sildenafil groups.
